Escherichia coli (E. coli) are gram-negative bacteria and are a type of faecal coliform bacteria commonly found in the intestines of animals and humans. They are invisible to the human eye and can only be seen using a microscope. Their growth can be seen as colonies on agar media under special conditions. Most E. coli do not cause illness but if a person becomes sick from E. coli, the primary site of infection is the gastrointestinal tract and symptoms can include nausea, vomiting, diarrhoea, and fever.
How to Test for Bacteria in Your Water
1) Get apparatus ready: test strip, reaction tube with lysate, pipette.
2) The reaction tube already contains 0.5ml lysate, Take sample water 0.1ml to reaction tube by pipette, Shake the mixture well.
3) Remove reagent test strip from foil (do not touch test pad). Immerse the reagent area (test pad) in water sample for 10 minutes.
Qualitative Test Results – 10 Minute Test (Positive or Negative)
Positive or Negative results can be observed after 10 minutes.
Positive (E. coli is present) = If the test strip changes to any shade of blue it indicates that E.coli is present. Continue to Semi-Quantitative Test.
Negative (E. coli is not present) = If the test strip remains white it indicates that E. coli is not present. Test is complete.
Semi-Quantitative Test Results – 60 Minute Test (Level of E. coli Present)
4) Immerse the reagent strip in the same test solution for an additional 60 minutes.
5) After 60 minutes, remove the test strip. Tap off excess water.
6) Compare your results to the color chart. Read the test results carefully in a well-lit area with the test pad held near the color chart.
7) Match the test strip colors with the corresponding colors on the chart and record your results.
